Hi
I want a user to enter a registration number into a field then validate agains a JSON endpoint from a third party system.
Is this possible with any of the current CB plugins? Or would a custom plugin be necessarry?

That's doable with CB Code Field. It provides PHP based validation support so you can implement whatever validation you need. So for example call an external API with cURL. Implements client side (ajax) and server side validation as well so it can live check inputs during registration or profile edit.
